I'm looking into a Silverado HD 2500 purchase.

I receive the gm supplier discount through work and the dealer is also offering a dealer promotion. Is it reasonable to expect those discounts could stack? I understand the supplier discount is paid from the manufacturer so I'm wondering whether dealer might have incentive to at least offer part of their dealer incentive...

Unfortunately it depends. There's so many ins and outs of where discounts come from. You'll have to ask the dealer.

They can, but don't always. Last time I was looking, they were willing to deal on 1500s but not on the 3/4 ton.
